This article details the development and synthesis of artificial mRNA using a novel chemical approach. This method uses 2-cyanoethoxymethyl as a protecting group and highly reactive phosphotriester pyrophosphorylating reagents to produce a 130-nucleotide mRNA encoding glucagon-like peptide-1 (GLP-1). The synthetic mRNA, capped and polyadenylated enzymatically, demonstrated biological activity by supporting protein synthesis in cell-free systems and in living cells. This research presents a scalable method for producing high-purity mRNA, offering potential for therapeutic applications and mRNA-based vaccines.
