    PrimeFlow RNA Assay

    PrimeFlow RNA Assay is based on the flow cytometry assay and in situ hybridization which combines the power of the single-cell resolution with branched-DNA signal amplification technology. This assay is capable of simultaneous detection of RNA within millions of cells at single-cell resolution. This assay could robustly detect up to three RNA transcripts at the single-cell level using a standard flow cytometer. Fluorescent in situ hybridization is an advanced tool that allows specific localization of RNA targets. Besides, branched-DNA technology can be used to amplify the RNA transcript to determine the differential mRNA expression within a heterogeneous population of cells. This assay can be used to detect mRNA, long non-coding RNA, microRNA, as well as viral RNA. The assay may be compatible with mammalian and non-mammalian cells. This RNA assay is combined with antibody staining to fill the gap of the understanding of the single-cell dynamics.

    Workflow of PrimeFlow RNA Assay

    In the PrimeFlow RNA assay workflow, cells are first labeled with cell-surface antibodies, initially fixed and permeabilized, and then labeled with intracellular antibodies. These cells are sequentially hybridized with probes specific for the RNA targets, and hybridized targets are detected after branched-DNA signal amplification. Individual probe pairs are designed to bind adjacent to the RNA transcript for bDNA signal amplification. Subsequently, the detection system is further magnified using amplifiers. Finally, multiple label probes with fluorescent dyes are hybridized on each corresponding amplifier. An optimally assembled branched-DNA complex can provide a theoretical 8,000- to 16,000-fold signal amplification of the targeted mRNA.
