Solid Lipid Nanoparticles (SLNs) in Delivery Systems

The use of lipid nanoparticles as drug delivery systems has been growing over recent decades. SLNs were developed at the beginning of the 1990s as an alternative carrier system to emulsions, liposomes, and polymeric nanoparticles as a colloidal carrier system for controlled drug delivery. SLNs are two-phase drug delivery systems consisting of a nanoscale drug-carrying solid phase composed of crystallized lipids at room temperature dispersed within an aqueous phase using high-pressure homogenization. The favorable physical and chemical properties, stability, good tolerability and biodegradability, and the possibility of large-scale production have demonstrated that SLN can be good vehicles for functional ingredients.

Carnauba Wax-based SLNs

Solid lipids utilized in SLN formulations include fatty acids, triglycerides, steroids, partial glycerides, and waxes (e.g., carnauba wax). Waxes are crystalline lipid material consisting of a complex mixture of long-chain fatty acids and fatty alcohols, hydrocarbons, aldehydes, and ketones. They show high availability at a low cost and great promises as constituents of delivery systems.

Carnauba wax is naturally obtained from the carnauba palm tree and is a solid material having high melting temperatures. Carnauba wax comprises C24 and C28 esters, C32 and C34 straight-chained primary alcohols, and hydroxy-fatty acids. It offers a wide variety of applications in the pharmaceutical industry. It has gained interest because of its biocompatibility, low cost, and sustained release of the drugs due to its high hydrophobicity. Other advantages include non-sensitivity to environmental stresses (pH and salt), the tunable microstructure, and no changes in structure during the homogenization process.
