Dimethyl Dioctadecyl Ammonium Bromide (DDAB), also known as Dioctadecyl Dimethyl Ammonium Bromide (DODAB), is a double-chained quaternary ammonium surfactant that can form unilamellar vesicles (ULVs) in water.

DDAB in mRNA Delivery

In recent years, studies have shown that permanently cationic, zwitterionic/anionic, or ionizable cationic lipids selectively induce mRNA delivery to the lung, spleen, or liver, respectively.

Dimethyl Dioctadecyl Ammonium Bromide (DDAB) is a common quaternary ammonium compound as well as a permanently cationic lipid. It can form a complex with mRNA and can also stimulate the innate immune response, thereby acting as an immune adjuvant for mRNA vaccines. Compared with DOTAP or DOPE, DDAB more stably binds with tRNA. In this case, DDAB can more stably promote the delivery of siRNA to tumor cells.

Using a film-ultrasonic method, solid lipid nanoparticles (SLNs) containing DDAB can be constructed and have been applied for miR-34a mimics delivery into the cancer stem cells to that increase the survival rate of CSC-bearing mice. As a cationic surfactant, DDAB presents genotoxic effects in mammalian cells at environmentally relevant concentrations, more research is still required to reduce the toxicity and improve the stability.
