High Pressure Homogenization

Instruction of High Pressure Homogenization

Hot homogenization and cold homogenization are two types of high pressure homogenization. With the advancement of nanotechnology, high-pressure homogenization technology has gradually broadened the scope of its pharmaceutical applications. Ease of production, short production time, uniform particle size reduction, and the avoidance of organic solvents make it a powerful and reliable technique that is widely used for the production of LNPs. On the other hand, the most fundamental advantage of high pressure homogenization is easy scale-up.

The process of this technology is more complicated. Briefly, high pressure homogenizers push a liquid with high pressure through a relatively narrow gap, then the fluid will accelerate at a very short distance to a very high speed. Based on this condition, the particles can be broken into submicron range by the high shear stress and cavitation forces. The homogenization technique has the advantage of easy scale-up, lack of organic solvents, and short production times. Based on the above advantages, the homogenization technique has been explored for the production of LNPs.

Applications of High Pressure Homogenization

In the past decade, much attention has been focused on LNPs which are emerging as a promising carrier for all administration routes due to their safety, small size, and high loading of lipophilic compounds. The high pressure homogenization method preparing LNPs has been explored for a wide range of delivery systems of numerous drugs, such as risperidone and clotrimazole.
